      DSS小鼠結(jié)腸炎造模解決方案

      小鼠模型是結(jié)腸炎造模常見模型,自從1985年首次采用葡聚糖硫酸鈉(dextran sulphate sodium, DSS)制備出倉鼠潰瘍性結(jié)腸炎模型后,已有大量數(shù)據(jù)證明DSS結(jié)腸炎模型的病因、臨床癥狀、病理改變及治療應(yīng)答均與人類潰瘍性結(jié)腸炎(ulcerative colitis,UC)相似。

       

      現(xiàn)行研究中常用的UC建模類型大約分為:自發(fā)模型、誘導模型(化學藥物誘導:TNBS三硝基苯磺酸,DSS葡聚糖硫酸鈉等、免疫學方法誘導、細胞移植誘導)以及基因修飾模型等。
       

      DSS造模優(yōu)勢

      葡聚糖硫酸鈉鹽(Dextran Sulfate Sodium Salt, DSS)是一種聚陰離子衍生物,其誘導結(jié)腸炎模型的機制雖仍未十分明確,但通常認為與巨噬細胞功能失調(diào)、腸道菌群失調(diào)、DSS對結(jié)腸上皮的毒性作用、細胞因子在DSS結(jié)腸炎模型的發(fā)病中起重要作用等機制有關(guān)。

       

      對比多種UC建模類型,DSS造模具有很大優(yōu)勢:

       

      1、癥狀與人類UC高度相似:可用于研究結(jié)腸炎發(fā)生機制和藥效研究;
      2、成模率高:自由飲用DSS水溶液,簡單易行、重復性強;

      DSS濃度:急性期模型通常為3-5%,慢性期模型通常為1-3%;可根據(jù)實驗需求及動物類型摸索濃度。該濃度為質(zhì)量體積比,使用飲用水配置后,0.22µM濾膜過濾除菌。

       

      數(shù)據(jù)記錄:每隔24h記錄小鼠體重、糞便粘稠度、糞便潛血等指標。

      急性DSS結(jié)腸炎模型

      Day1:稱重并標記各組C57BL/6小鼠。待造模小鼠飲用3-5% DSS水溶液,未處理組小鼠飲用正常水。
      Day3:給予待造模小鼠更換新鮮3-5%DSS飲用水。
      Day5:給予待造模小鼠更換新鮮3-5%DSS飲用水。
      Day8:給予待造模小鼠更換新鮮不含DSS的飲用水。

      Day1:稱重并標記各組C57BL/6小鼠。待造模小鼠飲用1-3% DSS水溶液,未處理組小鼠飲用正常水。
