VDR-Luc螢光素酶報(bào)告基因(報(bào)告基因質(zhì)粒)(VDR luciferase reporter plasmid)是翌圣生物自主研發(fā)的用于檢測(cè)VDR
轉(zhuǎn)錄活性水平為目的的報(bào)告基因。VDR(Vitamin D Receptor)在本質(zhì)上是類固醇受體超家族的成員之一。它在維持機(jī)體鈣、磷
代謝,調(diào)節(jié)細(xì)胞增殖、分化等方面起重要作用。
VDR報(bào)告基因主要應(yīng)用于檢測(cè)細(xì)胞VDRE信號(hào)通路中VDR的轉(zhuǎn)錄活性、藥物研究以及基因過表達(dá)和 RNAi 的表型分析等。
pVDR-Luc是翌圣生物改造后的哺乳動(dòng)物真核表達(dá)載體,在其多克隆位點(diǎn)插入了多個(gè)VDR結(jié)合位點(diǎn),可以高靈度地檢測(cè) VDR 的激活水平。同時(shí),對(duì)載體中預(yù)測(cè)出的其它轉(zhuǎn)錄因子以外的結(jié)合位點(diǎn)進(jìn)行了適當(dāng)?shù)耐蛔儯诒3衷泄δ懿蛔兊那闆r下,增加了質(zhì)粒的轉(zhuǎn)錄因子結(jié)合特異性。另外,由于質(zhì)粒體積減小,使得VDR報(bào)告基因質(zhì)粒更易于轉(zhuǎn)染。

使用說明
1)pVDR-Luc可以采用常規(guī)轉(zhuǎn)染方法轉(zhuǎn)染哺乳動(dòng)物細(xì)胞。用螢光素酶檢測(cè)試劑盒或雙螢光素酶檢測(cè)試劑盒進(jìn)行檢測(cè);
2)首次使用1 µg包裝的本產(chǎn)品時(shí),請(qǐng)先取少量本質(zhì)粒轉(zhuǎn)化大腸桿菌,進(jìn)行質(zhì)粒小量、中量或大量抽提后再用于后續(xù)用途。抽提獲得的質(zhì)??梢酝ㄟ^酶切電泳進(jìn)行鑒定,或通過測(cè)序進(jìn)行鑒定。
